1) SN1 and other Gln carriers involvement in molecular mechanisms related to astrocytic and neuronal functions in HE.

  • HE model implementation and characterization in SN1 KO mice

2) The SN1 transporter: activity, expression, structural modification, tissue and intracellular distribution.

  • Impact of ammonia on SN1 function in Xenopus levis oocytes or mammalian cell line transfected with SN1: two-electrode voltage clamp electrophysiology (TEVC),  molecular imaging and uptake assays

  • Tissue distribution of Gln/Glu and glutamine carriers in the brain of a) wild type + HE model and b) SN1 KO mice + HE model in vivo

3) Metabolism of Gln and its precursors/products in astrocytes-neuronal co-cultures derived from wt and/or SN1KO mice using the 13C NMR spectroscopy and mass spectrometry.
